IN T RO DU C T IO N One of the main challenges for a global lipid analysis (lipidomics) is the separation of the wide array of lipid species present in biological samples (Figure 1). Such a separation is not achievable using a single chromatographic dimension such as reversedor normal-phase separation methods. Normalphase UPLC separates lipid classes based on their polar head group, whereas reversed-phase separates lipids according to their acyl chain length and number of double bonds. Hydrophilic interaction chromatography (HILIC) separation has been proposed as an alternative to normal-phase separation, offering better MS compatibility and using less toxic solvents. Recently, a two-dimensional separation using HILIC and reversed-phase has been proposed to maximize the separation of the lipidome before MS detection.

Identification and separation of saxitoxins using hydrophilic interaction liquid chromatography coupled to traveling wave ion mobility-mass spectrometry.

The aim of this work was to develop a reliable and efficient analytical method to characterise and differentiate saxitoxin analogues (STX), including sulphated (gonyautoxins, GTX) and non-sulphated analogues. For this purpose, hydrophilic interaction liquid chromatography (HILIC) was used to separate sulphated analogues. We also resorted to ion mobility spectrometry to differentiate the STX ana...

Multiplexed ion mobility spectrometry-orthogonal time-of-flight mass spectrometry.

Ion mobility spectrometry (IMS) coupled to orthogonal time-of-flight mass spectrometry (TOF) has shown significant promise for the characterization of complex biological mixtures. The enormous complexity of biological samples (e.g., from proteomics) and the need for both biological and technical analysis replicates imposes major challenges for multidimensional separation platforms with regard t...

High resolution ion mobility-mass spectrometry for separation and identification of isomeric lipids.

Lipidomics is a particularly difficult analytical challenge due to the number and importance of isomeric species that are known or postulated in biological samples. Current separation and identification techniques are too often insufficiently powerful, slow or ambiguous. High resolution, low field ion mobility coupled to mass spectrometry is shown here to have sufficient performance to represen...

High-throughput shotgun lipidomics by quadrupole time-of-flight mass spectrometry.

Technological advances in mass spectrometry and meticulous method development have produced several shotgun lipidomic approaches capable of characterizing lipid species by direct analysis of total lipid extracts. Shotgun lipidomics by hybrid quadrupole time-of-flight mass spectrometry allows the absolute quantification of hundreds of molecular glycerophospholipid species, glycerolipid species, ...

QSRR models of veterinary drugs in milk in ultra-performance liquid chromatography coupled to time of flight mass spectrometry

The veterinary drugs residues are also important pollutants found in milk, since veterinary drugs are commonly used in cattle management. Considering the role of milk in human nutrition and its wide consumption throughout the world, it is very important to ensure the milk quality. A quantitative structure–retention relationship (QSRR) was developed using the partial least square (PLS), Kernel P...
